BABY HUG
BABY HUG Study Summary

BABY HUG is a randomized, double-blind multi-center clinical trial designed to enroll 200 patients between the ages of nine and seventeen months with sickle cell anemia. Each patient was assigned to treatment with hydroxyurea or placebo. The study is currently in a follow-up only phase; enrollment ended in September 2007. The primary analysis will be based on the detection of evidence of end organ damage (two organs of anticipated interest are the kidney and spleen) taking death, bone marrow transplantation and chronic transfusion into account as treatment failures. Treatment and follow-up will last two years. Double blinding of the assignment to hydroxyurea or placebo and medical treatment will be enhanced by the use of a Primary Endpoint Person at each Clinical Center to monitor local CBC results. Determination of the presence or absence of chronic end organ damage (either directly evidenced or inferred from surrogate measures) will be the responsibility of Endpoint Adjudication Panels composed of pediatric sub-specialists and sickle cell anemia expert(s) to be convened by the Medical Coordinating Center. The Endpoint Adjudication Panels will be provided complete clinical and laboratory documentation relevant to the classification of patient outcomes by the Medical Coordinating Center with attention to the blinding of personal identifying information and of treatment assignment information.

BABY HUG is conducted by the collaborating investigators of the Medical Coordinating Center (MCC), affiliated Core Laboratories, and 14 Clinical Centers; the National Heart, Lung, and Blood Institute (NHLBI) Project Office in the Division of Blood Diseases and Resources (Sickle Cell Scientific Research Group); the National Institute of Child Health and Human Development (NICHD) Program Office; and a Pharmacy Distribution Center. A Data and Safety Monitoring Board (DSMB) has been appointed by the NHLBI and serves in an advisory capacity to the Director of the NHLBI, providing recommendations based on study performance, interim and final analyses.
